Holders Hibernians will open their title defence with a clash with Challenge League side Gudja United as the draws of the Round of 32 of the cup competition were held on Wednesday.
However, the top clash of the round will see YoHealth Malta Premier League leaders Floriana take on Marsaxlokk.
Hibernians are going through a rather low-key campaign as they failed to qualify for the Top Six of the Opening Phase of the YoHealth Malta Premier League and could see the cup competition as a major objective to not only add more silverware but also qualify for European football.
The clash between Floriana and Marsaxlokk is set to be a fascinating encounter between two teams enjoying a positive start to the season.
So far, the Greens have enjoyed the upper hand over Marsaxlokk this season, as they have prevailed in both league clashes, winning with an identical 2-1 scoreline.
Ħamrun Spartans will face the winner of the Preliminary Round clash between Victoria Hotspurs and Xagħra United, while Mosta will take on Mqabba.
Sliema Wanderers face Swieqi United, while Birkirkara will play SK Victoria Wanderers.
Valletta face Pembroke Athleta while Tarxien Rainbows will face Gozitan side Għajnsielem.
Żabbar St Patrick will face the winner of the clash between Melita and Xewkija Tigers.
Naxxar Lions face Marsa, while Gżira United will be up against the winner of the tie between St Andrews and Vittoriosa Stars.
The FA Trophy Last 32 matches will be played between January 3 and 4.
